Influence of chemotherapy with 5-fluorouracil on the osseointegration of the installed implants in the rat tibia: biomechanical, histomorphometry, immunohistochemistry and microtomography analysis

Abstract

The aim of this study is to evaluate of the point of view tomographic, biomechanical, histomorphometric and immunohistochemical (TRAP and OPG markers), the influence of chemotherapy with 5-Fluorouracil (5-FU) during the process of implants osseointegration; and the influence of chemotherapy with 5-FU in the maturation of bone tissue in areas of osseointegrated implants placed in the rats tibia. Sixty animals will subjected to a surgical procedure for installing a titanium implant in the right tibia and a titanium implant in the left tibia. The animals will be randomly divided into four groups : group C15 (control) (n=20) - the animals will receive the first injection of saline solution 15 days after implant placement; 5FU/15 group (n=20), the animals will receive the first dose of 5-FU chemotherapy 15 days after implant placement; C30 group (control) (n=10) - the animals will receive the first injection with saline solution 30 days after implant placement; 5FU/30 group (n=10) , the animals will receive the first dose of 5-FU chemotherapy 30 days after implant placement. Ten animals of the C15 group and ten animals 5FU/15 group will be euthanized 30 days after implant placement, and ten animals in groups C15, 5FU/15, C30 and 5FU/30 be euthanized at 45 days, by means of an overdose anesthetic. The right and left tibias and will be collected and fixed in 4 % buffered formalin and 70% alcohol respectively. The specimens will be processed to perform the biomechanical, histomorphometry, immunohistochemistry and microtomography analysis. The normality of the data will be verified and statistical analysis will be realized with significance level of 5%. (AU)
