Abstract

Cardiac hypertrophy is a compensatory response that occurs in many cardiovascular diseases, and studies in isolated cardiomyocytes allow a more specific evaluation of the effects of hypertrophic stimuli and characterization of signaling pathways involved in cardiac hypertrophy development. Recently, some studies have shown that there are pro-inflammatory cytokines that play an important role in the development of cardiac hypertrophy. The Toll-like receptor, especially TLR2 and TLR4, receptors have also been described as participating in the hypertrophic process. However, knowledge in this area has become scarce. It is known that the signaling pathway NF-kB is classically activated by Toll-like receptors and is closely related to cardiac hypertrophy in different experimental models. This study aims to evaluate the participation of the signaling pathway NF-kB in the tropism of cardiomyocytes in vitro after activation of Toll-like receptors 2 and 4 for both receivers will be used primary cultures of neonatal rat cardiomyocytes divided into two major groups: 1 ) cardiomyocytes treated with agonists of Toll-like receptors 2:04 and 2) to cardiomyocytes treated with NF-kB RNAi or cardiomyocytes treated with NF-kB RNAi associated with agonists of Toll-like receptors 2 and 4. The study shows If promising since preliminary results show success in obtaining cultures of cardiomyocytes as well as in the analysis of gene and protein expression. (AU)
