Abstract

There is increasing need of nonhuman primates (NHPs) for biomedical research, which generates the need of efficient methods to ensure adequate supply. The use of ARTs have overcome many of the problems encountered. The rhesus monkey (Macaca mulatta) is one of the most commonly used NHP in research. Although, there have been many advances in the application of ARTs in this species, cryopreservation of sperm is still a major limitation. The objectives of this project are to test and compare different protocols for cryopreservation of sperm from rhesus monkeys and evaluate its efficacy by assessment of post-thaw survival and in vitro fertilization rates. Semen will be collected from five adult males by penile electroejaculation and frozen by two different methods (1) a slow freezing method and (2) a vitrification method. Oocytes will be collected from five adult females by laparoscopic follicular aspiration. Frozen-thawed sperm will be used for in vitro fertilization. Embryos will be examined daily until they reach the blastocyst stage. (AU)
