Abstract

The constant threats to non-human primate survival makes it necessary to intervene with Assisted Reproductive Technologies. A recent study assessed different cryopreservation protocols for Alouatta caraya semen. Although none of the results were satisfying, the best results were obtained using an egg yolk-based extender with 4% glycerol. The present project was elaborated based on the results of the latter and on other studies using different species. The study will be divided in two experiments: Experiment I - determination of glycerol concentration and freezing rate for Alouatta caraya semen; Experiment II - evaluation of the effects of adding DHA and vitamin E to Alouatta caraya semen. A total of seven males will be used, two from Fundação Parque Zoológico de São Paulo, São Paulo - SP and five from Centro Nacional de Primatas, Ananindeua - PA. In order to compare the different protocols, the following parameter will be assessed: sperm motility, sperm morphology, plasma membrane integrity, acrosome integrity, mitochondrial activity, susceptibility to oxidative stress (TBARS), and susceptibility of the chromatin to acid denaturation (SCSA). Additionally, Fourier Transform Infrared Spectroscopy (FTIR) will be used to obtain data regarding sperm composition before and after cryopreservation. Data will be analysed using SAS system for Windows.
