Abstract

In theory, the activity of the mitochondrial alternative oxidase (AOX) decreases ATP production by the respiratory chain of fruitflies that express this enzyme transgenically. Preliminary data show that carbohydrate poor diets have a significant impact on the development of AOX-expressing flies. To analyze in details this negative effect of AOX in conditions of nutritional stress, diverse diets will be tested, in comparison to the standard laboratory diet (rich in sugars, such as sucrose, glucose, corn flour, soy flour and treacle). The carbohydrate-restricted diets will contain only 0, 5 or 10% glucose, in addition to 3.5% yeast extract (source of amino acids and some fatty acids). Optimization of methods for analyzing the mitochondrial proteomics will be performed to provide a starting point to the investigation into the molecular nature of the developmental defect caused by AOX.
