Caries susceptibility of fluorosed teeth and fluoride effect on its control: in vitro and in situ study

Abstract

It was found in an in vitro study that enamel of teeth with mild/moderate fluorosis are less resistant to dental caries, but this result was obtain without the local effect of fluoride. Aim: To evaluate in vitro and in situ the local effect of fluoridated toothpaste on the reduction of the induced enamel demineralization. Methods: Enamel blocks will be obtained from teeth classified according to the Thylstrup & Fejerskov Index (TFI) in sound (TFI 0) and with fluorosis (TFI 1 to 4). Two studies will be conducted: one in vitro and another in situ, each containing two treatment groups: non-fluoridated placebo toothpaste and fluoridated toothpaste (1100 mg F/g). In the in vitro study, 20 enamel blocks from each TFI will be randomly allocated to one of the treatment groups. These blocks will be subjected to a pH-cycling model, and 2x/day enamel blocks will be treated according to the allocated group. At the end of this in vitro study, the hardness area in function of the lesion deep (”S) and the total fluoride concentration formed on enamel will be determined. The in situ study will be crossover and double-blind, conducted in two phases of 14 days with 15 volunteers. Each volunteer will use a palatal appliance with 10 enamel blocks (2 sound and 2 of each TFI). Enamel blocks will be treated 8x/day with a 20% sucrose solution and volunteers will brush their teeth and the appliance 2x/day with the correspondent toothpaste. On the 15th day, the fluoride concentration in the fluid and in the solid of biofilm will be determined. Also, the hardness area in function of the lesion deep (”S) and the total fluoride concentration formed on enamel will be determined. Expected Results: To extend the knowledge about caries susceptibility of fluorosed teeth and generate new data about the importance of local fluoride use for its control. (AU)
