Abstract

The plant embryogenesis is a highly complex process and organized in space and time, with a central role in the plant life cycle. The establishment and development of a new individual, the embryo by zygotic and somatic embryogenesis, has been used as a model for studies of development and differentiation in plants. Somatic embryogenesis, considered a biotechnological technique has been an alternative of clonal propagation for conservation and breeding of the species. Araucaria angustifolia is a native subtropical coniferous of Brazil that has recalcitrant seeds. Because of its economic importance, were heavily exploited over the years, currently classified as critically endangered species by IUCN. There are about twenty years, the research group of Laboratório de Biologia Celular de Plantas (BIOCEL) has used this system in plant physiological, biochemical and molecular studies, an integrative approach of zygotic and somatic embryogenesis. Different markers throughout embryogenesis has been investigated, however, carbohydrates and your metabolism, which admittedly are a key part of the processes of growth and plant development remains understudied. This project aims to investigate the role of carbohydrates and the integration of their metabolism with other markers, specifically ethylene and nitric oxide during embryogenesis zygotic and somatic A. angustifolia. For this, we used physiological, biochemical and molecular parameters such as carbohydrate profile (soluble carbohydrates, starch and cell wall), ethylene, nitric oxides, reactive oxygen species and differential expression of genes (according to the bank transcripts existing in BIOCEL).
