Abstract

This proposal is linked to the FAPESP research grant 2014/14318-5. In the lab we study the gene expression regulation of the segmentation cascade of Drosophila melanogaster. More precisely, we investigate the action of maternal and gap transcription factors in the regulation of the anterior most stripes of the pair-rule seven striped pattern. We test the hypothesis that there are several transcription factors acting in a redundant and/or combinatorial manner to avoid the expression of the enhancers responsible for these stripes in more anterior regions of the embryo, whereas these stripes are not formed. We accumulated genetic evidence that the gap gene tll could be one of this repressor genes. However, there are non evidence of the direct regulation of tll in the regulatory regions of putative stripes targets, which is the main goal of this project. More specifically, we propose to test in vitro the direct interaction between the transcription factor Tll with the regulatory element already characterized of the first stripe of the pair-rule gene even-skipped trough the eletrophoretic mobility shift assay (EMSA).
