Abstract

There are many research groups in the field of photodynamic therapy (PDT). However, some challenges were not yet overcome, such as results variability. The aim of this project is to advance the state of art of the technique, investigating the causes for the variability. It will be used a threshold dose distribution model to evaluate cell resistance to PDT. From quantitative comparisons of the distribution curves acquired in different treatment conditions, it will be investigated the relationship between PDT parameters and cell resistance. Conditions include different photosensitizer concentration (Photogem and Clorin e6) and incubation time and light doses in cell and microorganisms culture. After the PDT applications, cell viability will be analyzed by MTT and fluorescence analysis. The Grupo de Óptica develop research in photodynamic therapy since 1999 and it is a reference in the field. Not only in research, but also at applications: it develops the largest Brazilian clinical projects: Skin Cancer and Cervical Cancer. The facilities available at the group are high-technology laboratories for cell culture, microorganisms and in vivo experiments, among others. There are several equipment available for measurements, such as Confocal Microscopes and Spectrometers (UV-VIS and FTIR). Therefore, we believe that this project will expand the understanding of PDT mechanisms, which allows the advance of the technique. (AU)
