Abstract

The fungus Candida albicans, in situations of imune imbalance in the host, can become an opportunist pathogen and cause infections. Due to the resistence that microorganism has developed to conventional drugs, antimicrobial photodynamic therapy (aPDT) has shown promising results as an alternative therapy. The aim of this study will be evaluate the induction ablity that aPDT has in developing tolerant biofilms of C. albicans in association with curcumin (CUR - 40 µg/L) and LED light (18 J/cm2). Three strains of C. albicans will be used in the presente study (ATCC 90028 and susceptible and resistant isolates) and being planted on Sabourand Dextrose Agar plates to obtain the biofilms. The biofilms will be submmited to successive applications of aPDT in a sub-lethal dose with the values of colony-forming units being evaluated in each application of aPDT. There will be four study groups evaluated, namely: aPDT (PS+Light), PS in absence of light, only application of light and control (absence of PS and light). Once tolerance was induced, the biofilms will be cultivated again for characterization and quantification of the polymeric extracellular matrix (total biomass analysis, protein quantification, water soluble polysaccharide quantification, alcohol soluble polysaccharide analysis, eDNA analysis and confocal and laser microscopy analysis) and for evaluation of expression of genes related with the adhesion of C. albicans to the substrate and genes responsibles for development of biofilms (ALS1, ALS3, EAP1 e PHR1). To confirm the normality and homocedasticity of the data the Shapiro-Wilk test and Levene's test will be perfom, respectively. For inference analyses, the analysis of variance (ANOVA) and Kruskall-Wallis will be conduct with a significant level of 5% (±=0.05). (AU)
