Abstract

The study has the objective to evaluate the potential of an antimicrobial Photochemotherapy has to develop resistance of Candida albicans. Strains of resistant C. albicans (ATCC 96901) and susceptible (ATCC 90028) to fluconazole will be grown and exposed the antimicrobial Photochemotherapy (aPDT). After aPDT the colony forming units will be counted and survivors cells will be cultivated and exposed to the aPDT. This procedure will be repeated 10 times. The aPDT will be mediated by Photosensitizers (PSs) Curcumin and Photodithazine® associated to led light. Samples will be submitted to quantification of genetic expression by the technique of RT-qPCR, using specific primers for the genes related to the oxidative stress of the fungus, genes involved in the ergosterol synthesis, and the genes that express membrane transporter protein. Besides that, will also be evaluated the minimum inhibitory concentration (MIC) and the minimum fungicidal concentration (CFM), before and after the treatments to compare the fluconazole susceptibility between the strains (susceptible and resistant to fluconazole) of the groups evaluated. It will also be quantified the oxidative stress tolerance (ROS). The results will be evaluated by the most appropriate statistical method about the found data. (AU)
