Abstract

The rising resistance to conventional drugs from Candida species have directed the search for alternative methods of fungal elimination. In this context, antimicrobial photodynamic therapy (aPDT) peptides antimicrobial are inserted. The objective of this project is to evaluate the efficacy of the peptide LL-37 against C. Albicans strains susceptible and resistant to fluconazole and its effects when associated with curcumine-mediated photodynamic treatment. We will utilize two reference strains (susceptible and resistant to fluconazole) and two resistant clinical isolates. The curcumine and the LL-37 will be synthesized following the literature protocols. We will conduct a photochemical characterization of the curcumine in the presence and absence of LL-37, through spectrophotometry analysis. We will also conduct uptake analysis to check if the presence of LL-37 interferes on the internalization of curcumine in the fungal cells. We will estimate the minimal fungicidal concentration of LL-37 and conduct analysis of antimicrobial effect in planktonic samples of C. albicans strains in order to establish the concentration of curcumin and LL-37, with and without lighting, with application potential.Subsequently, we will investigate the antimicrobial effect on mono-species biofilm using curcumin, LL-37 and association of both, with and without lighting. The data will be analyzed with descriptive and inferential statistic, according to experimental design of each step. In all analyzes, the significance level will be 5%. (AU)
