GPR30 receptor involvement in the rapid effect of aldosterona in kidney

Abstract

Aldosterone is a steroidal hormone with an important role in renal electrolyte homeostasis. In vitro studies (in renal cells) have showed the involvement of GPR30 and ERK1/2 signaling pathway in aldosterone rapid action; however, similar investigations were not performed in renal tissue (in vivo). Thus, this project aims to: 1) detect in rat kidney (in vivo) the gene expression of GPR30 and the protein expression of ERK1/2 (total and phosphorylated) by PCR and Western blot, respectively; 2) study the rapid aldosterone effect (after 30 minutes) on the phosphorylation of ERK1/2 in renal cortex and medulla; and 3) verify the effect of the antagonism to the GPR30 receptor (using the drug G15) on this hormonal effect. The GPR30 has been considered the "new receptor" of aldosterone, and ERK1/2, an important signaling pathway for this mineralocorticoid; therefore, studying the involvement of GPR30 in the ERK1/2 pathway is relevant to clarify the mechanism of action of aldosterone.
