Molecular detection, quantitation, tissue distribution and determination of pathological changes of enteric viruses related to enteric disease in chickens.

Abstract

Enteric diseases cause significant economic problems, because the commitment of the digestive system of birds in the growth phase. Consequently, there is a reduction in weight gain, uneven growth of the lot, sometimes mortality. Among the enteric problems, relates to Runting Stunting Syndrome (RSS) considered one of the most important clinical manifestations of enteric viruses. The RSS is an enteric disease primarily characterized by diarrhea, stunted, loss of weight gain and mortality. Chicken parvovirus (ChPV), chicken astrovirus (CAstV) and avian nephritis virus (ANV) are associated with the RSS, identified in outbreaks worldwide. There is little information with respect to these three viruses being investigated as the cause of enteric problems, particularly diarrhea, symptom related with RSS in poultry. In Brazil, they were detected and isolated from healthy chickens and broilers or with enteric problems. Still do not know the involvement of these viruses as cause of enteric diseases. CAstV, ANV and ChPV isolated and stored in Avian Pathology Laboratory FMVZ- USP, will be inoculated in birds at one day of age in order to reproduce the syndrome. The birds will be divided into four (04) groups, one for each of the viruses named above and one which will serve as a negative control. This project aims to: 1) identifying and quantifying each studied virus by a real-time PCR reaction for each virus that will be standardized and validated; 2) to reproduce the runting and stunting syndrome by experimental infection using individual virus; 3) determining the macroscopic and microscopic changes induced by these viruses; 4) to detect the virus in infected cells by transmission electron microscopy.
