Abstract

Local anesthesia is commonly used to avoid pain during dental procedures. However it promotes anxiety and pain for most patients. Even though topical anesthetic are used to reduce discomfort associated to needle insertion and local anesthetic injection, the commercially available formulations presents reduced efficacy. The present study aims to evaluate in vitro mucoadhesive properties and permeation ability of biopolymeric films composed of chitosan and pectin containing lidocaine and prilocaine for topical anesthesia in Dentistry. It will be evaluated three different biopolymeric films containing 2.5% lidocaine and 2.5% prilocaine: 1) 2% chitosan; 2) 2% pectin; 3) 1% chitosan and 1% pectin. In vitro mucoadhesive properties will be evaluated in a texturometer and mucoadhesive strength (area under de curve of strength versus distance) and detachment force (force to detach the films from the mucosa); residence or mucoadhesion time and in vitro permeation across pig buccal epithelium in Franz type vertical diffusion cells. We expect that chitosan and pectin based films presents good mucoadhesive properties and allow permeation of local anesthetics across oral epithelium, being promising for topical anesthesia in Dentistry.
