Abstract

Senescence is associated to several metabolic disorders, including insulin resistance (IR). Physical exercise has an important role as non­pharmacological agent in the prevention and treatment of IR and the associated complications. Rho­kinase (Rock) has been postulated as an important protein that acts directly on the phosphorylation of IRS­1, contributing up to 50% of glucose uptake in skeletal muscle. Therefore, the objective of this study is to evaluate the role of physical exercise on Rock expression and glucose uptake in aged rats. Fischer 344 rats will be distributed into three groups: Sedentary Young (5 months) Middle Age (17 months) and Old (21 months). The MA and Old groups will be submitted to chronic (5 days) or acute swimming exercise (1 session) protocols, composed by 4-30 minutes swimming sessions with 5 minutes of rest between them. After the first session, a 1.5% of body weight workload will be attached to animal´s thorax for the last three bouts. Part of animals will submitted to the insulin tolerance test, another to the glucose tolerance test. Sixteen hours after the exercise session, the last animals will be euthanized for insulin signaling pathway and Rock metabolism analysis through immunoprecipitation, western blotting and Rock-activity assay techniques.
