Obtaining interspecific hybrids between guava (Psidium guajava L.) and araça (Psidium cattleianum ) through ovary culture

Abstract

The current scenario of Brazilian agriculture is being threatened by growing losses caused by the nematode Meloidogyne enterolobii that parasitizes the roots of guava, damaging all its development. A possible solution to this problem is to produce hybrids between guava and species of strawberry-guava (Psidium cattleianum) resistant to M. enterolobii. However, much of the interspecific crosses made so far do not show satisfactory results, which emphasizes the importance of the use of biotechnology tools such as embryo culture, to overcome this barrier to the success of hybridization. In front of this, three experiments will be conducted. The first is to test different genotypes of guava cross pollinated with strawberry-guava in situ, while in the second experiment pollination will be conducted in vitro. In both the aim is to evaluate the effects of different crosses in organogenesis / embryogenesis of the ovaries by evaluating the number of infected ovaries, oxidised, dead, early callus formation and embryos. In the third experiment, which aims to evaluate the effect of culture media on organogenesis / embryogenesis of the ovaries, four different culture mediums will be tested. Thus the objective of this research is to develop zygotic embryos resistant to nematode M. enterolobii from crosses between guava and strawberry-guava from ovaries cultured in vitro. Once the experiment involves the use of in vitro culture and ovaries recovery technique it can also be expected to obtain somatic embryos and / or haploid / haploid double, both processes that have not yet been reported in this genus.
