Abstract

The intestinal parasite Giardia lamblia, etiological agent of giardiasis, affects every year millions people all over the world. Most highlighted symptoms of giardiasis are diarrhea, flatulence, malabsorption syndrome and intense abdominal pain. The infection by Giardia lamblia starts with the ingestion of cysts (resistant and infective form of the parasite). Once in the stomach, the acid ambient combined with the presence of digestive enzymes lead the excystation of ingested cysts generating throphozoites (replicative form of the parasite). The adhesion of the parasite in the intestinal epithelium occurs after its passage through the mucus layer formed mainly by mucin family proteins highly glycosylated that are present in all intestine. It is known that some enteropathogenic organism, such the bacteria Escherichia coli, are able to modulate the expression of mucins. The aim of this project is to evaluate the expression of mucins in the host cell before and after the infection by trophozoites of Giardia lamblia using quantitative real time PCR analysis of mRNA, western blotting and Periodic Acid-Schiff stain.
