Abstract

Porcine circovirus type 2 (PCV2) are small, icosahedral, non-enveloped viruses containing a circular single-strand DNA genome, noncytopathogenic, which belong to the family Circoviridae. PCV2 is the causal agent of Post-weaning multisystemic wasting syndrome (PMWS), a great economic important disease worldwide distributed. The viral isolation of PCV2 in cell culture is difficult, laborious, and produces low viral titres. However, the preparation of cell cultures of swine testicle (ST) persistently infected with PCV2b, cultivated without performing passages, with high viral loads, and producing cytopathic effect, it was reported by our research group. As PCV2 infection is characterized by being persistent and causing severe immunosuppression, there is the need for more research to elucidate this important aspect of the pathogenesis of the disease. Therefore, the objective of this project is to perform the study of the transcriptional profile of persistently infected ST cells compared to the control cells, and animals experimentally infected with genotypes PCV2a, 2b, and 2c using RNA-seq technique (RNA sequencing). Then the genes that were differentially expressed will be analyzed by RT-qPCR "in vitro" (ST cells) and "in vivo" (animals natural and experimentally infected with PCV2). In addition, the next generation sequencing (NGS) will be used to sequence the entire viral genome of samples analyzed in this project. Thus, this research will be able provide new information regarding the pathogenesis of the disease, aiming to help in the prevention of this important viral disease. (AU)
