Evaluation of adaptation, empty spaces and intratubular penetration of an experimental selaer: analysis in laser scanning confocal microscopy

Abstract

The objective of the present study is to evaluate the adaptation, empty spaces and intratubular penetration of an endodontic cement. 30 unirradiculados teeth are to be used, which will be previously x-rayed for deletion of any anatomical changes. The crowns are removed by means of double-sided diamond blades coupled to low rotation, with the purpose of standardizing the specimens in 16 mm in length. The length of work will be established 1 mm short of the Summit. Initially the roots will be explored with K type 15 and instrumented file until the file Unicone 40.06 and irrigation will be held 2 mL with NaOCl to 2.5% every Exchange of instrument. At the end, 2 mL of EDTA will apply for 3 minutes and final irrigation with saline. Then will be divided randomly in 3 groups (n = 10): G1: AH Plus; G2: MTA FillApex; G3: Sealer Plus (experimental). All groups will be filled by single-cone technique, 40.06. Will be added to 0.1% of the cements dye rhodamine B for the purpose of viewing under a microscope. After obturadas, the roots will be stored in moisture for 72 hours for the final prey cements. The roots are sectioned transversally 2, 4 and 6 mm from the apex and analyzed in laser scanning confocal microscopy (540 to 590 nm), for evaluation of adaptation, empty cement penetration in the Dentinal tubules. To confront statistically the data obtained in each parameter evaluated, statistical programme will be used GraphPadPrism (La Joya, CA, USA). For the study of the comparison between the time and the experimental groups will be applied to analysis of variance (ANOVA) followed by the Tukey test for multiple comparisons among them being adopted 5% level of significance.
