Abstract

Gene-environment interaction (GxE) is being target as a novel avenue of exploration in the understanding of psychotic disorders (PD). Notably, childhood adversity (CA) is considered an important environmental risk factor in this context. Recently, the inflammatory hypothesis is being considered in the development of PD, with special attention to the participation of the cytokines IL-6, TNF-a and IL-10. Nevertheless, there is a lack of information regarding the association between CA, inflammatory profile and changes in cytokines gene expression (mRNA) in First Episode of Psychosis (FEP) patients. Our aims are: to determine the plasmatic concentration and the cytokines mRNA of IL-6, TNF-a and IL-10, in human samples and in the animal model of Schizophrenia; to investigate possible associations between plasmatic concentration and mRNA of the cytokines with a history of CA in FEP patients, compared to healthy controls and siblings; to investigate possible alterations in the animals' cytokines plasmatic concentration and mRNA (cerebral tissue and leucocytes); and to investigate possible similarities regarding animals and humans' results. Subjects (16-64 y/o) from Ribeirão Preto-SP and surround area were selected. Cytokines plasmatic concentration (ELISA) will be analysed of 166 FEP, 166 healthy controls, and 76 siblings. Cytokines gene expression will be analysed from a sub-sample of 30 Schizophrenia FEP, 30 healthy controls and 30 siblings. For the animal model, male Wistar rats (n=20) will be submitted to the post-weaning social isolation. Statistical analysis will be conducted according to the experimental protocol, and p<0,05 will be considered significant. (AU)
