Abstract

The blood vessels are present in all organs participating/regulating growth and homeostasis. They are formed, among other components, endothelial cells having specialized functions such as the ability to communicate to tissue / cells surrounding providing them with essential factors, such as cytokines, angiocina factors, chemokines and other metabolites in accordance with the needs. Endothelial cells found in blood vessels within the bone are able to regulate bone development. It is known that osteogenesis and angiogenesis processes are interrelated, endothelial cells and bone work together through a positive feedback mechanism to help each other to establish the proper development of the skeleton. Leukotrienes (LTs) are lipid mediators involved in the recruitment of leukocytes and in the regulation of cytokine synthesis. They are derived from arachidonic acid through 5-lipoxygenase pathway. The treatment with LTB4 induces angiogenesis by overexpression of vascular endothelial growth factor (VEGF). The individual responses of endothelial cells and bone LTB4 were demonstrated in studies, but as these cells respond together LTB4 has not been studied. In this study, we sought to investigate the potential contribution of LTB4 by bone cells in mediating VEGF endothelial cells which may play a role in osteoclastogenesis. To this end, primary bone vascular endothelial cells will be treated with the LTB4 and VEGF expression will be evaluated. Then the direct or indirect effect (dependent on osteoblasts) of VEGF secreted in osteoclastogenesis will be determined. We expect this project to understand the role of leukotrienes in angiogenesis linked to osteogenesis. (AU)
