Abstract

P. aeruginosa is responsible for severe periodontitis and from the supragingival plaque can be disseminated systemically. In infections, microorganisms may have strategic to inactivate the cells of the immune system and the use of medicinal plants can contribute to inhibit these actions, since components have proven effective, such as R. officinalis L. with dozens of biological activities. The aim of this study is to evaluate the action of R. officinalis L. extract on in vitro infection control induced by P. aeruginosa in RAW 264.7. The macrophages are cultured in 24-well plates for 24 h. Next will be exposed for 5 min to the plant extract. Penicillin-streptomycin will be used as control. The P. aeruginosa infection occurs for 30 min (multiplicity of infection of 1: 5). The cell culture will receive three doses of this product, one before and two after infection (4 and 8 h). At another moment, macrophages no longer receive prophylactic dose and only receive treatment. For immunological analysis, IL-1² levels of TNF-± and IL-10 supernatants are quantified and microbiological analysis will determine the concentration of colony forming units per milliliter (CFU / mL) obtained from macrophages. The cytotoxicity caused by infection will be evaluated by incorporation of neutral red method to lysosomes of viable cells. The data will be statistically analyzed by ANOVA and Tukey test (p d 0.05).
