Effect of incorporation of nanostructured silver vanadate on antimicrobial activity, flow ability, radiopacity and dental staining on endodontic sealers

Abstract

The antimicrobial activity, physical and mechanicals properties of dental materials can be improved by adding some agents, as based compounds silver ions associated with vanadate nanowires for its stabilization. The aim of this study is to evaluate in vitro mechanical (flowability), physical (dental staining and radiopacity) properties and antimicrobial activity of sealers incorporated with nanostructured silver vanadate. Initially the antimicrobial action of silver vanadate nanostructured will be evaluated against the species: Enterococcus faecalis, Pseudomonas aeruginosa and Escherichia coli. It will be used the sealers AH Plus, Sealapex, Endofil and Sealer 26, and produced 108 specimens for each, of dimensions of 4x3mm. The specimens will be divided into three groups according to the amount of silver vanadate (n = 27): control (no antimicrobial agent 0%), 5% and 10%. The antibacterial analysis will be performed through the agar diffusion method in triplicate and in three times: 2 days, 7 days and 21 days after incubation. The flow of the cement will be assessed by flow test, observing the interference of silver vanadate according to the concentration of the material in the cement. The specimens of each concentration will be placed on an occlusal film and exposed to a scale of aluminum with variable thickness of 5mm. The radiographs will be digitized using a scanner, in order to analyze its density, thereby comparing the various radiographic density cements and concentrations and different degrees of radiopacity thick aluminum. The radiographic images will be obtained using a digital X-ray machine Model Image with digital sensor Vistas can composed of phosphor plate and 16-bit system. Five measurements of each sample will be made. The average will be calculated. For staining of the tooth structure test, will be selected through a bank of teeth, upper and lower incisors 36 healthy, divided into 12 groups (n = 3), free of caries, restorations, cervical lesions and coronal discoloration. The teeth will be cleaned and sectioned in the coronal plane in the middle third of the root (in cross section), and apical segments will be removed. The pulps will be cut off and the pulp chambers instrumented. The channels will be irrigated with distilled water and dried with paper points. The cavities will be filled by the four sealers in three concentrations of nanostructured material: Control (0% silver vanadate), 5% silver vanadate and 10% silver vanadate. All teeth will be sealed with composite resin. and stored in a properly identified container containing distilled water at 37 ° C. The measurement of dental staining will be made before entering the sealers in the teeth by spectrophotometer. A new assessment of the color will be made with 1 week, 1 month, 3 months and 6 months. The results will be statistically analyzed in Minitab software version 15.1.1.0, with tests of normality and homogeneity to define the use of parametric or non-parametric analysis.
