Analysis of scaffolds influence of polycaprolactone (PCL) associated with nanohydroxyapatite (nHA), produced by rotary jet spinning in bone repair

Abstract

The bone repair process depends fracture type, mechanical stability and support as regenerative cells. Polycaprolactone (PCL) and a biocompatible synthetic polymer, biodegradable that in association with nanohidroxiapatita (nHAp) may give rise to a bioactive product, not toxic, low cost, with excellent mechanical properties and slow degradation. The rotary jet spinning and a biomaterial production technology, simple, inexpensive and allows fiber production micro / nano. Additionally this technique allows the incorporation of bioactive nanoparticles auch as a nHAp originated biomaterials make composite type (PCL + nHA). The objective this experiment was to evaluate will be production, characterization and bone formation capacity using PCL + nHAp scaffolds produced by rotary jet spinning technique. The scaffolds produced are identified by scanning electron microscopy (SEM). Subsequently, in vivo test it will be used 20 Wistar rats, male, from 90 days old, weighing 300g. The animals are divided randomly in 4 experimental groups (n = 5): G1-control (blood clot); G2 - PCL scaffold; G3 - PCL scaffold with 5% nHAp; G4 - PCL scaffold with 20% nHAp. It will be performed critical defect, with 3mm diameter, the tibia of animals, which will be filled with different scaffold. Control group (G1) will be performed defect and after a suture. All animals will be euthanized 30 days after the surgery. The area of bone repair in the right tibia will be assessed by means of radiographic analysis, histological analysis and histomorphometric. The bone repair area of left tibia wiil be submitted to flexural strength test. The data will be statistically analyzed, initially through the normal trial to the statistical test application suitable. The level of significance adopted will be the conventional value of 5%. (AU)
