Abstract

In this project, the immobilization of beta-galactosidase (EC 3.2.1.23) of Kluyveromyces lactis by the method of self-aggregation or cross-linked enzyme aggregates (CLEAs) will be held, in order to produce a stable derivative and with maximum retention of the enzyme catalytic activity that is useful for enzymatic synthesis of galactooligosaccharides (GOS), from lactose, and also to improve the stability and enzyme recovery process. Thus, immobilizations of ²-galactosidase will be performed varying the precipitant, the concentration of cross-linker and the concentration of feeder protein employed, evaluating the recovered activity of products, so that better conditions can be selected. Tests to determine the thermal stability of the derivatives compared to the free enzyme will also be carried out. Finally, it is intended to analyze the selectivity of the free enzyme and the produced biocatalysts, as well as their reuse capacity, aiming to use them in the production of GOS. (AU)
