Leptin envolviment in the alteration of body paramethers of mice submited to neonatal anoxia: analyses by the fos-ir and hypothamalic leptinergic neurons of the LeprBEGFP mice, after acute fasting

Abstract

Neonatal anoxia is a relevant clinical situation in Brazil, which has been studied in the neuroscience laboratory ICB-USP, with a model developed, adapted and already validated. Our previous studies have identified its deleterious effects on glial and neuronal populations of the hippocampal formation, neuronal death by apoptosis and necrosis, neurogenesis and volume changes in the hippocampus, loss to learning and spatial navigation of the animal, among other behavioral sequelae. It was also observed significant alteration of body parameters in animal anoxia relative to their controls, already evident in the first weeks of breastfeeding wich persisted into adulthood. The changed parameters of the body development sugests hypothalamic involvement and so the need to evaluate the hypothesis of the influence of leptin in these changes, what we plan to perform by the stereological analysis of colocalization of immunopositive cells to FOS in neurons that express the leptin receptor. In view of this specific population of neurons of LepR-IRES-Cre cruzados com tdTomato reporter mice (B6.Cg- Gt(ROSA)26Sortm9(CAG-tdTomato)Hze /J, JAXR mice) will be used, they are modified organisms whose cells expressing the leptin receptor are naturelly fluorescent under suitable illumination. The hypothalamus is structure directly related to the energy metabolism and to the growth of the organism; functions in which literature indicates involvement of leptin. The mice will be anoxiated in P3 and at 60 days they will be fasted for 18 hours, as a stimulus to the FOS expression. The glicemia will also be followed. This study aims to contribute to the understanding of the mechanisms that produce the developmental changes as well as to the develop strategies to minimize them and provide better living conditions for those affected by the neonatal anoxia.
