Effect of a new intracanal medication based on silver nanoparticles on fibroblasts

Abstract

The aim of this study is to evaluate the biocompatibility of a new intracanal medication with silver nanoparticles (SNs), developed by researchers from the University of Ribeirão Preto (UNAERP) on fibroblast cell cultures. For this purpose, fibroblast cells from 3T3 cell line will be plated at 20,000 cells per well in 24-well plates and allowed to adhere for 24 h. The cultures will be then exposed to SNs, Ca (OH) 2, AgNP + Ca (OH) 2 and Chlorhexidine (CHX) in three different concentrations (1x, 2.5x and 5x), based on the minimum inhibitory concentration (MIC) determined in a previous microbiological study of our research group. Cultures exposed only to vehicle (ultrapure water) will serve as control. The following parameters will be assessed: 1) cell viability at 1, 3 and 7 days; 2) cell morphology in 3 days; 3) oxidative stress (NO production) in 1 and 3 days, 4) genotoxicity in 3 days and 5) gene expression of proinflammatory cytokines: tumor necrosis factor alpha (TNF-±) and interleukin 1 (IL-1) and type I collagen. Quantitative data will be submitted to normality test and equality of variances test for selecting the appropriate statistical test (alpha = 5%). (AU)
