Swimming and anthocyanins in liver and pancreatic histology of Wistar rats treated with high calorie diet

Abstract

The excess body fat can affect the liver and pancreatic morphology in animals that consume industrialized diets. Thus, we suggest this project in order to evaluate the effects of aerobic exercise and dry extract of red-orange Citrus sinensis (L.) Osbeck on the liver and pancreatic histology of male Wistar rats treated with a high-calorie diet. The total of 84 animals will be allocated to the maintenance of animal facilities and divided into seven groups, which will consist in the commercial diet (control); high-calorie diet; control diet associated with red-orange; calorie diet and red-orange; control diet and swimming; calorie diet and swimming, and high-calorie diet + swimming + red-orange. Since the beginning of the experiment, the rats are weighed weekly. The groups that practice physical training will undergo two weeks of adaptation until they reach 30 minutes of daily activity. At the beginning and the end of the experiment will be evaluated body composition of all groups in Hologic, Discovery Wi equipment, which will consist of fat percentage (%), amount of body fat (g), and amount of lean body mass (g). In the same age, the animals are weighed, euthanized for collecting the liver and pancreas, which will also be weighed to calculate the relative weight (%). Samples of these bodies will be fixed in 4% formalin, processed for histological routine, and stained with Hematoxylin-Eosin (HE). The histological slides will be analyzed in a photomicroscopy for morphometry evaluation. In the liver, the hepatic lobe area and the number of hepatocytes will be measured, and, in the pancreas, the area of pancreatic acinar and the pancreatic islets. It will be investigated if the red-orange has preserver action in the morphology of these organs even in animals that eat high-calorie diets.
