Abstract

Amniotic and allantoic fluids composition shows about the fetal metabolism. The analysis of these fluids indicates the fetal physiology, placental functions and changes that can affect pregnancy in this period. Thus, the proteomics has been an important tool to elucidate physiological and pathological modifications in fluids and cells, can be utilized to study the fetal maturation. In this way, the present project aims to study the protein profile modifications of the fetal fluids during the buffalo gestation. Fetal fluids will be collected from 3 gestation periods (1/3 initial, middle and final), the fetuses will be measured, sexed and the lung maturity will be evaluated by the Clements test (surfactant amounts). The total protein concentration will be measured and the protein digestion will be performed with trypsin. Then, the digested samples will be evaluated by mass spectrometry (MS). After the analysis, the gene ontology will be annotated and interactomes will be constructed, respecting the gestational ages. We will intend to identify proteins related to development and fetal maturity.
