Abstract

Among the most important challenges relevant to aquaculture, the occurrence of viral infectious diseases is one which needs our attention, having devastating effects. During the past two decades, the vírus genus Ranavirus (family Iridoviridae) have been associated with mortality events in amphibians, fish (wild and cultivated) and reptiles, with significant ecological and economic losses to the aquaculture. However, the knowledge of the pathogeneses of ranavirose disease, referring to the immune defense mechanisms of fish, relative to ranavirus, remains a challenge. In this context, this project looks to investigate the initial immune response by a brazilian strain of frog vírus type 3 (FV3), in fish source cells, and verify the possible activation of the apoptoses mechanisms. For this to be possible, CHSE-214 cells, derived from embryological epithelial cells of King Salmon (Oncorhynchus tschawhycha), and RTS11 cells, derived from monocyte/ macrophage Rainbow Trout (Oncorhynchus mykiss), will be infected by FV3, in order to measure the gene expression of genes of the innate imune sytem (IL-1², TNF-±, IL-10, IL-8, Mx, IFN-± e MHC I), by qPCR, during virus infection. In addition, the FV3 capacity to induce apoptotic in CHSE-214 e RTS11cells, will be examined by DNA fragmentation detection, phosphatidylserine exposure and assessment of cellular morphology. We believe that the results of this study will contribute to elucidate the innate immune response triggered by FV3 fish, and collaborate with subsidies for the characterization of potential immunogens.
