Abstract

The oocyte in vitro maturation (IVM) is one of the main bottlenecks in vitro production of bovine embryos (IVP). The existing protocols for IVM do not reflect the physiological conditions and determine mismatch between nuclear and cytoplasmic maturation of the oocyte, thus compromising their fertility. Oocytes undergoing IVM are from follicles in different stages of development and this heterogeneity hinders the establishment of ideal growing conditions. The aim of this project to develop a new system for IVM, follicular system, which includes a strategy for synchronization of follicular activity in donor cow in order to obtain homogeneous population of oocytes, as well as sequential cropping system adjusted to the maturation phase of oocytes recovered. The experiments included in the project aimed at developing / processing protocols for ovarian synchronization donor cows and for crops sequences which mimic the conditions intrafollicular before (pre-IVM phase) and during ovulation (IVM phase).
