Abstract

The physical exercise is highly recommended to type 1 diabetic (T1-D) patients, due to its benefits to this population, e.g., reduction in insulin needs, improvement of cardiorespiratory fitness, macronutrients metabolism, and skeletal-muscle function. However, the optimal exercise prescription to T1-D remains a challenge, since it is difficult to adjust the insulin dosage with the exercise. Moreover, the hypoglycemic risk is remarkable increased during and post exercise. The mainly contributors to this paradigm are: Maintenance of hyperinsulinemia, glycogen storage reduction, diminished hepatic glucose production, and glucagon secretion below the demand imposed by the hypoglycemic state. Thus, therapeutic strategies focuses on glucagon rise are promising. In this sense, dipeptidil peptidase-IV (DPP-IV) inhibition seems to be a potential candidate to prevent hypoglycemia risk in DT-1, during and post exercise, since that drug increased glucagon secretion during hyperinsulinemic-hipoglicemic clamp. Keeping it in mind, we aim to explore the effect of vildagliptina (a DPP-IV inhibitor) upon glucagon secretion and the possible efficacy of that drug in counteract the hypoglycemic events in T1-D C57BL-6 mice, during and post exercise.
