Evaluation of the influence of chemotherapy with cisplatin or 5-Fluorouracil over the peri-implant bone repair process of osseointegrated implants inserted in tibiae of rats modified or not by nicotine

Abstract

The purpose of the present research will be to evaluate the influence of chemotherapy over bone repair around osseointegrated titanium implants inserted in tibiae of rats exposed or not to nicotine. One hundred eighty male rats (Wistar) will be divided in six groups (n=30). At day "0" the animals will receive an implant (4x2.2mm - SLA) into their right and left tibial proximal metaphysis. During the 30 days before and after the surgical intervention the animals will receive two injections via subcutaneously of 3mg/kg nicotine hemi sulfate or saline solution daily, via dorsal and with 12 hours interval between them. After 5 days cessation, the protocol for chemotherapy will consist on administration via intraperitoneal of two doses of saline solution, 5-Fluorouracil ( first dose 60 mg/kg; second, 40 mg/kg) or cisplatin (first dose 5 mg/kg; second, 2,5 mg/kg) at 35 and 37 days postoperative. SS (SHAM): will receive applications of saline solution and simulation of chemotherapy with saline solution. SS-5FU: will receive saline solution subcutaneously and chemotherapy with 5-Fluorouracil. SS-CIS: will receive saline solution subcutaneously and chemotherapy with cisplatin. NIC-SS: will receive applications of nicotine and simulation of chemotherapy with saline solution. NIC-5FU: will receive applications of nicotine and chemotherapy with 5-Fluorouracil. NIC-CIS: will receive applications of nicotine and chemotherapy with Cisplatin. The euthanasia will be at 60, 65 and 95 days postoperatively. The collected tibiae will be submitted to conventional histological processing for histologic, bone area measurement and immunohistochemistry for detection of BMP2, RUNX-2, OCN and TRAP analyses and to undecalcified ground-section processing for bone/implant contact and ultrastructural analyses. After analysis of normality the data will be submitted to statistical analysis though the most appropriate tests (p<0,05). (AU)
