Conjugation of superparamagnetic nanoparticles of iron oxide and curcumin: association of photodynamic therapy and magnetic hyperthermia

Abstract

The incidence of cutaneous infections caused by bacterial agents resistant to conventional antibiotic treatment has become a major public health problem. Some alternative therapeutical approaches have been proposed as the Photodynamic Therapy, which involves the generation of reactive oxygen species through the combination of light and a photosensitizer, and Magnetic Hyperthermia, which promotes the increase of the temperature of an anatomical region by subjecting a superparamagnetic material to an alternating magnetic field (AMF). In order to promote the combination of these two approaches in a single material, it is proposed in this project the preparation of a ferrofluid based on superparamagnetic maghemite nanoparticles and curcumin. In the synthesis will be used different proportions of curcumin in relation to maghemite, in order to investigate the influence of curcumin in the colloidal stability of ferrofluid and in the proposed applications. This investigation and characterization of the material will be carried out using X-ray diffraction, electron transmission microscopy, zeta potential, small-angle X-ray scattering, Fourie transform infrared spectroscopy, differential scanning calorimetry, thermogravimetry, SQUID magnetometry, heating assays with application of a AMF and fluorescence spectroscopy. The use of this material in photodynamic therapy and magnetic hyperthermia will be evaluated through microbiological assays with planktonic Staphylococcus aureus bacteria, as well as the synergism of the two therapeutic approaches will be investigated through the simultaneous use of a light source and a AMF. In addition, the use of nanoparticles of maguemite as photosensitizers in photodynamic therapy will also be evaluated. (AU)
