Abstract

In this project, we will continue to collect human adipocyte samples and generate RNA-seq and histone modification ChIP-seq libraries to assess genome-wide transcriptional and epigenomic states. We have identified key cis-regulatory elements with altered activity in the IR state and have determined the candidate transcription factors (TFs) that bind them. We will validate the importance of candidate TFs by deleting or overexpressing them in both mouse and human cultured adipocytes and assess insulin sensitivity by glucose uptake assays and insulin signaling.
