Abstract

Antibodies (Immunoglobulins) are glicoproteins employed in the analytical, therapeutic, diagnostic, biosensor, and proteomic areas. Purification of IgG molecules is critical, since homogenous antibody preparations are usually required for these applications. Also, the need for large doses of antibodies for therapeutic treatments led researchers develop new chromatographic techniques for antibodies separation. In the last years, one the developed in this area is the monolithic beds as stationary phase in chromatography. For their macroporous structure, the monolithic beds have suitable properties for purification of macromolecules, like macroporous structure, low resistance to flow through the column and mass transfer governed mainly by convection. This project aims to develop an adsorbent based on acrylamide, allyl diglidyl ether, and alginate, prepared by the technique of cryo-polymerization, to be used as a monolithic stationary phase for the adsorption of human IgG. The pseudobiospecific ligand ortho-phosphotyrosine (OPT) will be immobilized in this cryogel, since our research group demonstrated that this ligand selectively adsorbed IgG from human plasma. The cryogels prepared based on polyacrylamide and alginate will be characterized as porosity, morphology, and water absorption. The adsorption capacity of IgG from human serum will be evaluated and the portion of IgG (Fab or Fc) adsorbed on the immobilized ligand will be evaluated. (AU)
