The influence of beta 2-adrenoceptor in satellite cell function

Abstract

Skeletal muscle tissue has a remarkable regenerative capacity due to the presence of muscle stem cells called satellite cells. The regeneration of injured myofibers includes the inflammatory process installation, the satellite cell activation and subsequent proliferation. Subsequently, a small population of satellite cells returns to the quiescence state and most of them differentiate to recover from previously injured muscle tissue. Although the morphological alterations that occur during muscle regeneration are well known, the molecular alterations involved in this process are not totally clarified. In this context, initial studies using treatment of synthetic beta2-adrenergic agonists in animals suggested that the beta2-adrenoceptor could be a possible regulator of the muscle regenerative process, because they demonstrated that this treatment was able to anticipate the satellite cell proliferation and enhance the structure and function of regenerating skeletal muscles. Our group also demonstrated that the beta2-adrenoceptor contributes, mainly to initial stages of muscle regeneration, being particularly involved in the regulation of macrophage recruitment to the regenerating muscle possibly through TGF-beta receptor I (Tbeta R-I) e smad2/3 activation. Furthermore, the beta2-adrenoceptor is involved in collagen remodeling during muscle regeneration, through reduction of matrix metallopeptidase 9 (MMP-9) activity. Although some works demonstrated that the beta2-adrenoceptor is important to muscle regeneration, the role of the beta2-adrenoceptor in the context of satellite cell function is not well understood. Therefore, we decided to investigate the influence of the beta2-adrenoceptor in satellite cell function. (AU)
