Despite the breakthrough in the treatment of some types of Cancer, the search for new therapeutic targets and antitumor therapy strategies to prevent metastasis, relapse and resistance to therapy is mandatory. The present proposal aims to define the role of sphingosine kinase 2 and SET in signaling by sphingolipids in oral Cancer. We will carry out studies on human cell derived from tumor and non-tumor. The different methods selected to achieve the proposed goal are (i) analysis of sphingolipids by mass spectrometry; (ii) post-translational modifications in proteins by Western blots; (iii) protein distribution by immunohistochemistry, immunofluorescence or Western blotting; (iv) flow cytometry to analyze cell cycle and cell viability, quantification of RNA by real-time PCR, interference RNA, invasion and clonogenic assays. These studies, in addition to the formation of xenograft tumors in nude mice will increase our knowledge on sphingolipid signaling and Head and Neck Cancer, as well as open new therapeutic opportunities against this type of Cancer.
