Abstract

Ascidians (Tunicata), or sea squirts, are benthic marine invertebrates widely investigated as a source of new chemical structures with pharmacological potential. More than 1000 molecules were described from ascidians so far. However, the production of most of these metabolites was latter attributed to associated-bacteria, which live a syntrophic association with the tunicate. This relationship between associated-bacteria and their host still is a universe very poorly understood, and efforts on the investigation of this interaction should be encouraged. There are many published works in high impact journal investigating the microbiota associated to ascidians, however, most of them collect the animal and process it entirely, using this mixture as a source of microbes and metabolites. Nonetheless, ascidians are organisms with organized systems, including ganglion, stomach, intestine, heart, a tunic that covers the body, among others. Considering that, some questions arises: (i) how does the interaction of ascidians and their microbiota influences the secondary metabolism? (ii) how is the microbiota distributed through the ascidians body? To get some insights regarding these issues, we would like to investigate the metabolic and microbial profile of different organs and tissues from ascidians body by employing mass spectrometry based metabolomic approaches. We believe that investigations that cover different aspects of this research field remains of extremely importance for science. Mass spectrometry (MS) is a versatile and very sensitive technique that when coupled to chromatography allows the analyses and detection of metabolites in complex samples, even metabolites at low concentration, very important characteristics to investigate biological samples. The MS data will be analyzed by Molecular network, an online molecular tool, able to grouping the compounds from the entire data set by structural similarity, besides helping in the identification of known compounds in the sample. Moreover, here, we are proposing the use of such MS data in order to construct a 3D volume model to better visualize the distribution of metabolites in ascidians body. Such approach, recently developed by Pieter Dorrestein's group (data not published, in revision at Nature), allows the visualization of molecular distribution through an organism's body, considering separately each part, laying a foundation for studies of metabolism and microbial-host-environment interaction.
