Proteomic analysis of tears in patients with oral potentially malignant lesions and oral squamous cell carcinoma

Abstract

The World Health Organization considers oral leukoplakia (LO) as white plaques on the oral mucosa that can not be clinically and histopathologically defined as any other lesion, with a considerable potential for malignancy. Proliferative verrucous leukoplakia (LVP) is a type of lesion that more frequently affects female patients (4: 1), older than 70 years, is not associated with smoking or alcoholism and has high rates of recurrence and malignancy. These lesions may lead to Oral Squamous Cell Carcinoma (OSCC). CEC is the most common malignant neoplasm in the oral cavity and will affect more than 350,000 people worldwide by the year 2018. In this way, understanding the etiopathogenesis of this condition and developing techniques to anticipate the diagnosis of malignization are essential for intervention at lower stages aggressive diseases. Thus, the identification of common proteins between the different stages of these lesions and in oral cancer can help to understand the development of these conditions and to reveal signatures markers of progression, indicating its potential for malignant transformation. Recent studies have shown lacrimal fluid as a promising source of cancer-associated proteins, especially breast cancer. Therefore, the objective of this study is to characterize the proteomic profile of the lacrimal fluids of patients with LO, LVP and CEC by means of mass spectrometry-based proteomics and to identify common proteins that may be linked to the malignant transformation potential of oral OSCC. Proteins will be analyzed by discovery-based proteomics, and those that are common between the different stages of progression, and not present in the control, will be considered potential and will be evaluated by immunohistochemistry in tissue obtained from group-independent biopsy of patients. The aim of this project is to indicate signatures of candidate proteins associated with the potential for malignant lesions and to anticipate the decisions regarding the treatment. (AU)
