Abstract

Direct pulp capping is a therapy with a limited number of clinical options, which is based on materials that cause coagulation necrosis of pulp tissue, followed by tissue regeneration mediated by the regenerative potential of dental pulp stem cells (DPSCs). The aim of this research project is to develop macro-porous chitosan scaffolds capable to activate reparative dentinogenesis process by DPSCs for dentin repair. The scaffold development will be based on the association of different tissue engineering techniques with the aim of obtaining an interconnected macro-porous network to allow cell infiltration. Following formulation and preparation technique selection, the scaffolds will be submitted to a hydroxyapatite biomineralization process by incubation in simulated body fluid. The DPSCs will be seeded on selected scaffolds, and the following parameters will be evaluated: chemotaxis, cell viability, cell proliferation, cell migration inside scaffold structure, odontogenic differentiation and mineralized matrix deposition. Numerical data will be subjected to statistical analysis. (AU)
