Abstract

Orthodontic Tooth Movement (OTM) results from an intense bone remodeling in a sterile environment. The mechanical stimulus applied to the tooth generates a cascade of physical and biological events in the periodontium, forming a tension and a pressure area in the alveolus of the tooth, under the action of osteoblasts and osteoclasts respectively. These cell types demand a high energy rate during differentiation and especially during their action, leading to the activation of AMPK. AMPK is a heterotrimeric serine / threonine protein kinase complex, composed by three subunits: alpha (±), beta (²) and gamma (³). This protein plays a central role in energy homeostasis, acting in situations of energy deficit (both at reduced ATP or increased AMP levels), in order to inhibit energy consuming processes and stimulate the producers. Several studies have evaluated the role of AMPK in osteoclasts and osteoblasts individually. Considering that the osteoblast is an important controller of osteoclast differentiation and function, the present project aims to study the role of AMPK on bone remodeling in an OTM experimental model. For this purpose, AMPK conditional knockout mice for osteoclasts (CtskCre AMPKflox/flox) and osteoblasts (Col2a1Cre AMPKflox/flox) will be used. A NiTi spring will be bonded in the upper right first molar, anchored to the incisors. The following parameters will be evaluated: distance reached by OTM by histomorphometry (H & E); number of osteoblasts (Masson's Trichrome) and osteoclasts (TRAP reaction); maxillary bone microarchitecture by MicroCT; expression of osteoclasts and osteoblasts markers in the alveolar bone by qPCR and Western Blot; seric inflammatory mediators by ELISA.
