Abstract

The ArtinM lectin, obtained from the saline extract of Artocarpus heterophyllus seed, recognizes Man± 1-3 (Man± 1-6) Man in N-glycans core. This mechanism is used by ArtinM to induce the activation of neutrophils and macrophages by TLR2/CD14 recognition, and T cell activation through CD3 interaction. The immunomodulatory activity of ArtinM on these cells demonstrates a protective effect against intracellular pathogen infections. We have reported the effect of ArtinM on B cell activation in a TLR2/CD14-independent-manner. This current proposal will improve the understanding of the effect of ArtinM on B cells in the context of immunomodulatory activity triggered by lectin. This proposal also includes the evaluation of the effect of ArtinM on NK cells, which acts on immune response against fungal infections, and we know that the in vivo administration of ArtinM protects against paracoccidioidomycosis and candidiasis. Therefore, we will investigate the in vitro assessment of the direct effect of ArtinM on B and NK cells by measuring the production of cytokines and chemokines, the expression on cell surface of activator molecules, the relative expression of cell differentiation markers and the secretion of cytotoxic granules. In addition, we will evaluate ArtinM-glycoprotein interaction in B and NK cells using immunoblot assay and functional competition assay. The in vivo effect of ArtinM on B and NK cells will be evaluated using spleen cells from naive mice that received ArtinM. Our proposal will demonstrate the involvement of B and NK cells in the immunomodulatory activity of ArtinM. (AU)
