Protein supplementation on longevity and gene expression related to the detoxification system of Apis mellifera bees exposed to the lethal dose of fipronil inseticide

Abstract

The goal of the present proposal is to evaluate the effect of protein supplementation on longevity and in detoxification related gene expression of Apis mellifera bees exposed to the lethal dose of the pesticide Fipronil. For this, five colonies will receive protein supplementation (25% crude protein offered weekly in the amount of 200g / colony) during the off-season (july), and five colonies will not receive any supplementation (control). Four weeks after the beginning of protein supplementation, open brood combs will be collected in the colonies and kept in an incubator until the birth of the bees. The newly-emerged bees will be distributed in experimental cages where they will continue to receive protein diet during the first six days of life or not; and later they will be challenged by ingestion with the LD50% of the insecticide fipronil (0.009 ± 0.003 ¼g / bee) for evaluation of total longevity and expression of the Cytochrome P450 306A1 and Glutationa-s-trasnferase 1 gene. Gene expression will be evaluated by Real-Time PCR, 12 and 24 hours after exposure of bees to LD50% of fipronil. The results will be evaluated using Analysis of Variance (ANOVA) followed by the Dunn test and will be considered statistically different when P <0.05.
