In vivo study of inflammatory response, maturation of collagen fibers, and expression of mineralization markers in pulp tissue of rats submitted to bleaching therapy using violet LED

Abstract

The in-office dental bleaching it has been strongly associated with sensitivity. In order to minimize these effects, the use of violet light-emitting diodes (LEDs), with or without of bleaching gels, has been proposed and represents today a new approach of treatment used by professionals. However, this recent technology still presents questions not clarified in relation to the biological alterations caused in the dental tissues after their application. Therefore, the objective of this study will be to evaluate the influence of bleaching therapy using Violet LED, with or without the use of bleaching gel based on 17,5% hydrogen peroxide (HP), on inflammation and expression of inflammatory immunomarkers and of mineralization in pulp tissue, and maturation of collagen fibers of dentin and pulp. 80 rats will initially be distributed in two experimental lots, and the experimental groups being randomly divided according to the treatments to be performed in the upper molars, on the right or left side: Control - no treatment, HP - 1 application of 30 minutes of 17,5% HP, LED - 1 application of 20 minutes of Violet LED, and HP+LED - application of HP concomitant to Violet LED. The animals will be euthanized immediately, and 7, 15 and 30 days after bleaching (n = 10). After processing the specimens, the first molar on each side will be analyzed histopathologically to assess the degree of inflammation and expression of proinflammatory markers interleukin (IL)-17 and IL-23 and of mineralization marker, the osteocalcin (OCN). The level of maturation of collagen fibers of dentin and pulp will also be evaluated. After the data collection, these will be subjected to appropriate statistical tests for each type of analysis.
