Abstract

Ratoon stunt (RSD), caused by the fastidious bacterium Leifsonia xyli subsp. xyli (Lxx), is considered the main disease of sugarcane due to difficulty of identification in field, which hampers its control. Information on disease impact on sugarcane yield is limited in Brazil, even though it is disseminated in all states and host cultivars. The main measures of RSD control are diagnostic assays and thermotherapy. The serology technology is employed in routine diagnostic analysis in Brazil due to logistic reasons to transport vascular fluid juice, higher capacity to examine a larger number of samples, and accessible cost to the sugarcane industry. Nevertheless, the detection level of Lxx is1 x 10 6 CFU/mL and effect on yield depletion below this detection threshold is unknown. Thermotherapy also has some aspects to be clarified for a better RSD control. For instance, the combination 52°C/30min is still employed in the Country because larger number of propagating materials can be treated in comparison to 50°C/2h, although it did not eliminate the pathogen when canes had high Lxx titer. Therefore, knowing the consequences of this failure is paramount to increase efficiency of RSD control. In case of a significant yield depletion occurs, an early detection technology capable of identifying the remaining Lxx is needed. Based on the above considerations, the present work aims to answer the following questions: a) What is the yield depletion resulting of the possible failure of the serology diagnostic test and the shortthermotherapy?, b) What is level of Lxx elimination by thermotherapy across Lxx titers?, c) How early can the pathogen not detected in serology diagnostic test or noteliminated by thermotherapy be detected by serology and molecular diagnostic tests?
