Influence of flunixin meglumine and firocoxib on LUTH phase and embryo receptor pregnancy rate in buffaloes

Abstract

Administration of non-steroidal anti-inflammatory drugs (NSAIDs) at the time of embryo transfer is usually employed to inhibit prostaglandin F2± (PGF2±) production in an attempt to prevent luteolysis. The aim of the present study was to evaluate the influence of flunixin meglumine (FM) and Firocoxib (FIRO), prostaglandin synthesis inhibitors, on luteal phase duration and progesterone (P4) plasma concentrations in buffalo females. The effect of the application of these drugs, at the time of embryo innovation, on the pregnancy rate of recipients and to identify the possible mechanism by which FM and FIRO positively affect the pregnancy rate of these animals. For this purpose, two experiments will be performed, the first using 40 adult crossbred buffaloes, synchronized by the administration of two doses of sodium cloprostenol (Sincrocio® 5, 500 g, intramuscular), with an interval of 11 days between applications. estrus considered as day 0. After 6 days of estrus detection, females will be submitted to reproductive tract ultrasound evaluation to determine the corpus luteum (CL) characteristics and divided into 4 groups: G1 (control, n = 10) without manipulation. uterine, G2 (uterine manipulation, n = 10), G3 (manipulation + FM application, n = 10) and G4 (manipulation + FIRO application, n = 10). All animals will be submitted to ultrasound examinations and hormonal measurements between D6 and the end of the luteal phase (D12, D18 and D21) to determine the P4 plasma concentration profile and CL functional duration. In the second experiment will be used 18 females divided randomly into three groups: G1 (control): where 6 animals will undergo reproductive tract manipulation similar to embryo innovation on day 6 of the estrous cycle; G2: manipulation group + FM application (1.1 mg / kg); G3: handling group + FIRO application (0.1 mg/kg). For the statistical analysis of the evaluated characteristics will be used the statistical package Statistical Analysis System, at a significance level of 95% (p <0.05). (AU)
