Thyroid hormones (HT), thyroxine (T4) and triiodothyronine (T3) are involved in the regulation of energy expenditure and thermogenesis, but alteration in HT levels is often associated with changes in body weight. The change in body weight is related to increased oxidative stress, which is a manifestation of metabolic imbalance related to obesity. A primary intracellular target for oxidative stress, which may be responsible for the adipocyte-induced obesity-induced tissue damage or altered HT levels, is represented by the mitochondria that synthesize the decoupling proteins, with emphasis on uncoupling Protein1 (UCP1). Antioxidants, such as beta carotene, have the ability to slow the rate of oxidation through one or more mechanisms, among them the inhibition of free radicals that are generated in obesity. The present work aims to evaluate whether T3 and beta carotene have an effect on oxidative stress and lipid profile in 3T3-L1 adipocytes. For this, a 3T3-L1 adipocyte culture will be used, which will be treated with Beta carotene (BC) - 20¼M ² 'carotene, for 72 hours; Triiodothyronine (T3) - 100nM T3, for 24h; BC + T3 - 20¼M ² 'carotene, for 72 hours and subsequent incubation with 100nM T3 for 24 hours and Control (C), absence of treatment. After the treatments will be measured the intracellular lipid accumulation, triglycerides, oxidative stress products through the dosages of maladonoid and carbonylation, as well as to verify the protein synthesis of UCP1 by Western Blot. The use of antioxidants may alleviate oxidative stress caused by alterations in T3 levels or lipid accumulation in adipocytes, and may be used as adjunctive therapy to other procedures, a fact with which this work will contribute.
