Abstract

Pesticides are used both at home and in agriculture to combat pests. However, its toxic effects are not limited only to organisms that cause damage to crops, but also affect soil, water, air and other organisms that initially would not be their target. Among pesticides, organophosphates (OFs) including chlorpyrifos, are among the most used, their mechanism of action being the inhibition of the enzyme acetylcholinesterase (AChE) resulting in toxic effects in several systems. On the other hand, some bacteria have the ability to degrade pesticides OFs and it is believed that this ability is due to the production of the enzyme alkaline phosphatase, including the probiotic Lactobacillus rhamnosus.Objectives. To evaluate the efficacy of the probiotic Lactobacillus rhamnosus against the neuroblocking effect of chlorpyrifos in neuromuscular preparation (explained: the inhibition of AChE by the pesticide leads to an increase in acetylcholine (ACh) in the synaptic cleft, the excess of which causes desensitization of nicotinic synaptic receptors, which in turn leads to a blockage of the contractile response).Methods. The experiments will be performed on phrenic nerve-diaphragm preparations of mice (ex vivo), using a conventional myographic technique. Dose-response curve will be performed with the pesticide, with the selection of concentration for further study with the probiotic, in 3 experimental models: pre-treatment with probiotic; post treatment with probiotic and pre incubation probiotic + pesticide 30 min before adding to the bath containing the preparation. From the better protective pharmacological model will be determined the cholinesterase in aliquots collected from the bath and the muscles will be analyzed histologically. Expected results. To evaluate the contribution of the probiotic against the chlorpiriphos effect using a functional experimental model. (AU)
